The long-running ITV1 programme will be transformed into a two-hour theatre performance at Salford's Lowry Theatre in August before touring the country as part of its landmark birthday plans.
Although it will not contain any past or present members of cast, the show will take fans through the major storylines of the past 7,000 episodes.
Executive producer Kieran Roberts has promised fans of the show will not be disappointed with the dramatic plotline - which he insists will also be filled with "lots of laughter".
I'm thrilled at the prospect of bringing to life on stage all the great stories and characters of fifty years of 'Coronation Street', and I'm sure the play will be a real treat for Corrie fans as well as a great night out in the theatre
He told the BBC: "The very heart of British television has been beating with the pulse of 'Coronation Street' for fifty years. That's a lot of memories, a lot of drama, a lot of poignancy and a lot of laughter.
"I'm thrilled at the prospect of bringing to life on stage all the great stories and characters of fifty years of 'Coronation Street', and I'm sure the play will be a real treat for Corrie fans as well as a great night out in the theatre".
The play will be written by the show's long-term scriptwriter Jonathan Harvey - who will write in parts for some of the programme's best-loved stars, including Curly Watts, Bet Lynch and Hilda Ogden.
Meanwhile, veteran actress June Whitfield has signed up to star in the soap.
The 84-year-old actress has been confirmed to play new character May, who turns up in Weatherfield to attend the funeral of her old friend Blanche Hunt - who was played by the late actress Maggie Jones - in May.
