Blackbird will see the actress work with director Patrik-Ian Polk for the first time, while Isaiah Washington is also on board.
Blackbird follows a teenage boy whose realisation he is gay rocks his small, religiously conservative town in Mississippi.
At the same time, the youth's parents are going through divorce and his sister goes missing.
Mo'Nique is set to play the mother of the teenager: who will be played by newcomer Julian Walker.
Mo'Nique will also serve as a producer on the film alongside husband Sidney Hicks.
Despite having a long and varied career, it really was Precious that put Mo'Nique on the map as she went on to dominate the awards season that year.
Blackbird is released later this year.
